چکیده

• On theoretical grounds, the high level of circulating antiplasmin in man was considered an objection to the clinical use of plasmin. Norman postulated that normal human plasma contains at least 30 times the amount of antiplasmin necessary to neutralize the plasmin which could be formed by activating all circulating plasminogen. Nevertheless, in controlled clinical studies" plasmin proved to be an effective clot-dissolving agent in a variety of thrombo-embolic conditions. Since the therapeutic dose of plasmin in these studies was insufficient to neutralize all circulating inhibitors, its mode of action remained an intriguing question. Several investigators have reported the high affinity of plasmin to fibrin" and suggested that the enzyme, once in high concentration on the clot, is not susceptible to inhibition by circulating antiplasmin. Others suggested that plasmin is not responsible for thrombolysis per se, but attributed therapeutic effects to its ability to activate plasminogen trapped into the clot. It was found' in our laboratory, that after an infusion of labeled plasmin into dogs, fibrinolytic activity of the blood rapidly decreases, while radioactivity persists in the circulation for a long time. The question arose whether in these experiments, circulating radioactivity does not represent a plasmin-

Suppression of argatroban-induced endogenous thrombolysis by PKSI-527, and antibodies to TPA and UPA, evaluated in a rat arterial thrombolysis model.

We have previously confirmed, using a rat mesenteric arteriole thrombolysis model, that thrombin inhibition induces endogenous thrombolysis in vivo. In addition, we have shown that thrombin-activatable fibrinolysis inhibitor (TAFI) plays a role in the down regulation of endogenous thrombolysis. However, the mechanism of endogenous thrombolysis or spontaneous plasmin generation in vivo remains u...

Plasminogen Activation With Streptokinase

Background. Platelet activation occurs in vivo during pharmacologic thrombolysis and may contribute to recurrent thrombosis. Plasmin does not directly activate platelets except at high concentrations; thus, the mechanisms for platelet activation during thrombolysis remain undefined. Increases in thrombin activity also occur in patients treated with fibrinolytic agents and may contribute to acti...

New approaches to thrombolytic therapy.

Tissue-type plasminogen activator (t-PA), purified from the culture fluid of a stable human melanoma cell line, is a serine protease, different from urokinase, with a molecular weight of about 70,000. It is composed of one polypeptide chain, which is converted to a two-chain molecule by limited plasmic action. Activation of plasminogen to plasmin occurs by cleavage of the Arg 560-Val 561 peptid...

Thrombolysis with plasmin: implications for stroke treatment.

Plasmin is a direct-acting thrombolytic agent with a striking hemostatic safety advantage over plasminogen activators in animal models of thrombolysis and bleeding. In contradistinction to plasminogen activators, which risk bleeding at any effective thrombolytic dose, plasmin is tolerated without bleeding at several-fold higher amounts than those needed for thrombolysis. Plasmin has been safe i...

Thrombolysis of occluded femoropopliteal graft with locally delivered human plasmin.

INTRODUCTION Acute lower limb ischemia results from thrombosis or embolization of diseased native artery or previously implanted bypass graft. When this occurs, several options are available to restore blood flow: catheter-directed thrombolysis, mechanical thrombectomy or open surgery. Fundamental reasons to apply percutaneous interventions are avoiding open procedures in high risk patients, an...
